2008 VL250.
This has happened twice recently.
I can generally only hear a loud clicking noise (starter motor?) and have to put choke on max & wide open throttle to make it even go.
Battery? Should I check it with my multimeter?

The clicking is probably the starter motor solenoid.
You are right - check the battery; it might be time to get a new one

Fuel pump? (if they have one) Based on my bike makes quite a ticking type noise if it hasn't been used for a while which I assumed was something to do with fuel. Plus you say it goes with full choke and throttle, fuel again?

Loud clicking noise ? Like constant clic clic clic clic clic, more than likely battery or connections !
Or one loud click ! Could mean starter bendick(sp) spring not engaging
A/ check battery connections !
B/ charge battery !
C/ buy a battery minder if you dont ride often enough (NB starting bike in shed does not count, you need to do at least 2-5 k to recharge what you use during a start up)
D/ Last resort if it's the starter or Solanoid and symptoms continue, seek medical advice !
