The Big Lebowski is up there. It's not laugh-a-minute but it has a nice overall effect.

Even though it lost it's way a bit, 'Porky's' made me laugh harder than I've ever laughed at a movie. The scene where the fella stuck his dick through the changing room wall and the aftermath of that in the principal's office. I literally could not catch my breath I was laughing so hard and the tears were streaming down my face.
'The Life of Brian' had some good moments throughout too. Particularly...

    CROWD: Ohhhhh!

BOB: Well, what about Wodewick, then?

CROWD: Yes! Welease Wodewick! Welease Wodewick!

PILATE: Centuwion, why do they titter so?

CENTURION: Just some, uh, Jewish joke, sir.

    PILATE: Are they... wagging me?

    CENTURION: Oh, no, sir!

    GUARD #3: chuckling

PILATE: Vewy well. I shall welease... Wodewick!

    CROWD: laughing

CENTURION: Sir, we don't have a 'Roderick' either.

    PILATE: No 'Woger'? No 'Wodewick'?

    CENTURION: Sorry, sir.

    PILATE: Who is this 'Wod'--

    GUARD #1: chuckle

PILATE: Who is the 'Wodewick' to whom you wefer?

    BOB: He's a wobber!

    CROWD: laughing

I'm going for the lesser known brother and toting The Meaning of Life... technically you could class it as a group of sketches, but you'd be wrong... Full of priceless gems, but my what has to be my favourite piece of comedic cinema has to be the following:

    and

Failing that i'll plump for Uncle Buck... gotta be one of the best family comedy's ever... and John Candy was, hmmmmm what word to use... sublime fits i guess... classic...
    I didn't think!!! I experimented!!!
