I don't worry about checking front/rear wheel alignment with string, I just make sure that I have turned the adjusters exactly the same number of part turns from the same reference point on the swing arm on each side.

I used this doc, from pyrocam, helped me the first time I tried tightening my chain.
Just make sure you also remove any lock pins before you undo the nuts.

I just looked around the back of my bike, grabbed the provided under-seat toolkit, looked carefully at the nuts, and adjusted them until the chain was properly tightened... Oh I LOVE BIKES SOOOOOO MUCH, so nice and easy to fix (so far...)
